Titulaire d'une maîtrise de droit public, il est marié à Marella Moretti, directrice des affaires financières de Fiat finances et services.
Yves Pozzo di Borgo est Inspecteur général de l'administration de l'éducation nationale et de l'enseignement supérieur.
En 1998, il est élu adjoint au maire du VIIe arrondissement, Martine Aurillac. En mars 2001, il est élu Conseiller de Paris.
Il est élu sénateur de Paris en septembre 2004. Il était le seul représentant UDF de Paris au Parlement français avant de rejoindre le Nouveau Centre .
En 2007, à l'élection présidentielle, il soutient François Bayrou au premier tour, qu'il a parrainé, puis Nicolas Sarkozy.
En septembre 2011, il est réélu sénateur de Paris.
Il est membre de la Commission des affaires étrangères et de la défense du Sénat. Il est l'un des représentants du Parlement français au Conseil de l'Europe.
